ABOUT THE SPEAKER
Danny Hillis - Computer theorist
Inventor, scientist, author, engineer -- over his broad career, Danny Hillis has turned his ever-searching brain on an array of subjects, with surprising results.

Why you should listen

Danny Hillis is an inventor, scientist, author and engineer. While completing his doctorate at MIT, he pioneered the concept of parallel computers that is now the basis for graphics processors and cloud computing. He holds more than 300 US patents, covering parallel computers, disk arrays, forgery prevention methods, various electronic and mechanical devices, and the pinch-to-zoom display interface. He has recently been working on problems in medicine as well. He is also the designer of a 10,000-year mechanical clock, and he gave a TED Talk in 1994 that is practically prophetic. Throughout his career, Hillis has worked at places like Disney, and now MIT and Applied Invention, always looking for the next fascinating problem.

Danny Hillis: Understanding cancer through proteomics

Danny Hillis: Înţelegerea cancerului prin proteomică

Filmed:

00:53
First, I'm going to try to give you a differentdiferit perspectiveperspectivă of genomicsgenomica.
13

3000

01:09
about how to go about treatingtratare cancercancer.
19
54000
2000
despre cum am putea trata cancerul.
01:11
So let me startstart with genomicsgenomica.
20
56000
2000
Aşadar voi începe cu genomica.
01:13
It is the hotFierbinte topicsubiect.
21
58000
2000
Este un subiect fierbinte.
01:15
It is the placeloc where we're learningînvăţare the mostcel mai.
22
60000
2000
01:17
This is the great frontierfrontiera.
23
62000
2000
Aceasta este marea frontiera.
01:19
But it has its limitationslimitări.
24
64000
3000
Dar are limitările sale.
01:22
And in particularspecial, you've probablyprobabil all heardauzit the analogyanalogie
25
67000
3000
Şi anume, probabil că toţi aţi auzit analogia
01:25
that the genomegenomului is like the blueprintBlueprint of your bodycorp,
26
70000
3000
conform căreia genomul este harta corpului uman.
01:28
and if that were only trueAdevărat, it would be great,
27
73000
2000
Dacă ar fi corect, ar fi perfect,
01:30
but it's not.
28
75000
2000
dar nu este.
01:32
It's like the partspărți listlistă of your bodycorp.
29
77000
2000
01:34
It doesn't say how things are connectedconectat,
30
79000
2000
Această hartă nu ne arată conexiunile,
01:36
what causescauze what and so on.
31
81000
3000
ce cauzează ce şi aşa mai departe.
01:39
So if I can make an analogyanalogie,
32
84000
2000
Deci dacă aş putea face o analogie,
01:41
let's say that you were tryingîncercat to tell the differencediferență
33
86000
2000
să spunem că am încerca să arătăm diferenţa
01:43
betweenîntre a good restaurantrestaurant, a healthysănătos restaurantrestaurant
34
88000
3000
dintre un restaurant bun, un restaurant sănătos
01:46
and a sickbolnav restaurantrestaurant,
35
91000
2000
si un restaurant prost
01:48
and all you had was the listlistă of ingredientsingrediente
36
93000
2000
01:50
that they had in theiral lor lardercămară.
37
95000
3000
pe care le au în cămară.
01:53
So it mightar putea be that, if you wenta mers to a FrenchFranceză restaurantrestaurant
38
98000
3000
01:56
and you lookedprivit throughprin it and you foundgăsite
39
101000
2000
şi uitându-te pe listă, ai afla
01:58
they only had margarinemargarina and they didn't have butterunt,
40
103000
2000
02:00
you could say, "AhAh, I see what's wronggresit with them.
41
105000
2000
02:02
I can make them healthysănătos."
42
107000
2000
Îi pot face sănătoşi."
02:04
And there probablyprobabil are specialspecial casescazuri of that.
43
109000
2000
02:06
You could certainlycu siguranță tell the differencediferență
44
111000
2000
Cu siguranţă că am putea observa diferenţa
02:08
betweenîntre a ChineseChineză restaurantrestaurant and a FrenchFranceză restaurantrestaurant
45
113000
2000
dintre un restaurant chinezesc şi unul franţuzesc
02:10
by what they had in a lardercămară.
46
115000
2000
prin ceea ce au în cămară.
02:12
So the listlistă of ingredientsingrediente does tell you something,
47
117000
3000
Aşadar lista ingredientelor ne indică ceva anume,
02:15
and sometimesuneori it tellsspune you something that's wronggresit.
48
120000
3000
02:19
If they have tonstone of saltsare,
49
124000
2000
Dacă deţin tone de sare,
02:21
you mightar putea guessghici they're usingutilizând too much saltsare, or something like that.
50
126000
3000
am putea bănui că folosesc prea multă sare, sau ceva asemănător.
02:24
But it's limitedlimitat,
51
129000
2000
Dar este o abordare limitată,
02:26
because really to know if it's a healthysănătos restaurantrestaurant,
52
131000
2000
02:28
you need to tastegust the foodalimente, you need to know what goesmerge on in the kitchenbucătărie,
53
133000
3000
02:31
you need the productprodus of all of those ingredientsingrediente.
54
136000
3000
02:34
So if I look at a personpersoană
55
139000
2000
02:36
and I look at a person'spersoane genomegenomului, it's the samela fel thing.
56
141000
3000
02:39
The partparte of the genomegenomului that we can readcitit
57
144000
2000
02:41
is the listlistă of ingredientsingrediente.
58
146000
2000
este lista ingredientelor.
02:43
And so indeedintr-adevar,
59
148000
2000
Şi astfel, într-adevăr
02:45
there are timesori when we can find ingredientsingrediente
60
150000
2000
sunt cazuri în care putem identifica ingredientele
02:47
that [are] badrău.
61
152000
2000
care sunt dăunătoare.
02:49
CysticChistica fibrosisfibroza is an exampleexemplu of a diseaseboală
62
154000
2000
Fibroza cistică este un exemplu de boală
02:51
where you just have a badrău ingredientingredient and you have a diseaseboală,
63
156000
3000
pentru care ştim ingredientul dăunător
02:54
and we can actuallyde fapt make a directdirect correspondencecorespondenţa
64
159000
3000
02:57
betweenîntre the ingredientingredient and the diseaseboală.
65
162000
3000
dintre ingredient şi boală.
03:00
But mostcel mai things, you really have to know what's going on in the kitchenbucătărie,
66
165000
3000
03:03
because, mostlyMai ales, sickbolnav people used to be healthysănătos people --
67
168000
2000
03:05
they have the samela fel genomegenomului.
68
170000
2000
ele au acelaşi genom.
03:07
So the genomegenomului really tellsspune you much more
69
172000
2000
Deci genomul indică mult mai multe
03:09
about predispositionpredispoziţie.
70
174000
2000
despre predispoziţie.
03:11
So what you can tell
71
176000
2000
Astfel, ce se poate evidenţia
03:13
is you can tell the differencediferență betweenîntre an AsianAsiatice personpersoană and a EuropeanEuropene personpersoană
72
178000
2000
03:15
by looking at theiral lor ingredientsingrediente listlistă.
73
180000
2000
doar prin compararea listei de ingrediente.
03:17
But you really for the mostcel mai partparte can't tell the differencediferență
74
182000
3000
03:20
betweenîntre a healthysănătos personpersoană and a sickbolnav personpersoană --
75
185000
3000
dintre o persoana sănătoasă şi una bolnavă --
03:23
exceptcu exceptia in some of these specialspecial casescazuri.
76
188000
2000
cu excepţia unor cazuri speciale.
03:25
So why all the bigmare dealafacere
77
190000
2000
Aşadar de ce se face atâta caz
03:27
about geneticsgenetică?
78
192000
2000
despre genetică?
03:29
Well first of all,
79
194000
2000
Ei bine, în primul rând,
03:31
it's because we can readcitit it, whichcare is fantasticfantastic.
80
196000
3000
03:34
It is very usefulutil in certainanumit circumstancesîmprejurări.
81
199000
3000
Este foarte util în anumite circumstanţe.
03:37
It's alsode asemenea the great theoreticalteoretic triumphTriumf
82
202000
3000
De asemenea, reprezintă şi un triumf teoretic
03:40
of biologybiologie.
83
205000
2000
al biologiei.
03:42
It's the one theoryteorie
84
207000
2000
Este singura teorie
03:44
that the biologistsbiologi ever really got right.
85
209000
2000
pe care biologii chiar au nimerit-o corect.
03:46
It's fundamentalfundamental to DarwinDarwin
86
211000
2000
Fundamentează teoria lui Darwin
03:48
and MendelMendel and so on.
87
213000
2000
03:50
And so it's the one thing where they predicteda prezis a theoreticalteoretic constructconstrui.
88
215000
3000
03:54
So MendelMendel had this ideaidee of a genegenă
89
219000
2000
Mendel a avut ideea de genă,
03:56
as an abstractabstract thing,
90
221000
3000
dar la nivel abstract.
03:59
and DarwinDarwin builtconstruit a wholeîntreg theoryteorie
91
224000
2000
Şi Darwin a construit o întreagă teorie
04:01
that dependeddepindea on them existingexistent,
92
226000
2000
care depindea de existenţa ei.
04:03
and then WatsonWatson and CrickCrick
93
228000
2000
Apoi Watson şi Crick
04:05
actuallyde fapt lookedprivit and foundgăsite one.
94
230000
2000
au cercetat şi chiar au găsit una.
04:07
So this happensse întâmplă in physicsfizică all the time.
95
232000
2000
Aşa se întâmplă mereu în fizică.
04:09
You predictprezice a blacknegru holegaură,
96
234000
2000
Previzionezi o gaură neagră,
04:11
and you look out the telescopetelescop and there it is, just like you said.
97
236000
3000
04:14
But it rarelyrar happensse întâmplă in biologybiologie.
98
239000
2000
04:16
So this great triumphTriumf -- it's so good,
99
241000
3000
04:19
there's almostaproape a religiousreligios experienceexperienţă
100
244000
2000
aproape că este o experienţă religioasă
04:21
in biologybiologie.
101
246000
2000
în biologie.
04:23
And DarwinianDarwinista evolutionevoluţie
102
248000
2000
Teoria evoluţiei a lui Darwin
04:25
is really the coremiez theoryteorie.
103
250000
3000
este chiar nucleul teoriei.
04:30
So the other reasonmotiv it's been very popularpopular
104
255000
2000
Alt motiv pentru care este foarte populară
04:32
is because we can measuremăsura it, it's digitaldigital.
105
257000
3000
04:35
And in factfapt,
106
260000
2000
De fapt,
04:37
thanksMulțumiri to KaryKary MullisMullis,
107
262000
2000
mulţumită lui Kary Mullis,
04:39
you can basicallype scurt measuremăsura your genomegenomului in your kitchenbucătărie
108
264000
4000
îţi poţi măsura practic genomul în bucătăria ta
04:43
with a fewpuțini extrasuplimentar ingredientsingrediente.
109
268000
3000
cu câteva ingrediente în plus.
04:46
So for instanceinstanță, by measuringmăsurare the genomegenomului,
110
271000
3000
De exemplu, prin măsurarea genomului,
04:49
we'vene-am learnedînvățat a lot about how we're relatedlegate de to other kindstipuri of animalsanimale
111
274000
4000
am învăţat foarte multe despre înrudirea cu alte animale
04:53
by the closenessapropierea of our genomegenomului,
112
278000
3000
datorită genoamelor asemnănătoare,
04:56
or how we're relatedlegate de to eachfiecare other -- the familyfamilie treecopac,
113
281000
3000
04:59
or the treecopac of life.
114
284000
2000
sau copacul vieţii.
05:01
There's a hugeimens amountCantitate of informationinformație about the geneticsgenetică
115
286000
3000
Există o cantitate mare de informaţii despre genetică
05:04
just by comparingcompararea the geneticgenetic similaritysimilitudine.
116
289000
3000
doar prin compararea similitudinilor genetice.
05:07
Now of coursecurs, in medicalmedical applicationcerere,
117
292000
2000
Acum, bineînţeles, ca aplicaţie în medicină,
05:09
that is very usefulutil
118
294000
2000
este foarte util
05:11
because it's the samela fel kinddrăguț of informationinformație
119
296000
3000
deoarece este acelaşi tip de informaţie
05:14
that the doctordoctor getsdevine from your familyfamilie medicalmedical historyistorie --
120
299000
3000
05:17
exceptcu exceptia probablyprobabil,
121
302000
2000
doar că probabil
05:19
your genomegenomului knowsștie much more about your medicalmedical historyistorie than you do.
122
304000
3000
genomul tău ştie mai multe despre istoricul tău medical decât tine.
05:22
And so by readingcitind the genomegenomului,
123
307000
2000
Aşadar prin citirea genomului,
05:24
we can find out much more about your familyfamilie than you probablyprobabil know.
124
309000
3000
putem afla mult mai multe despre familia ta decât probabil ştii tu.
05:27
And so we can discoverdescoperi things
125
312000
2000
Putem descoperi lucruri,
05:29
that probablyprobabil you could have foundgăsite
126
314000
2000
05:31
by looking at enoughdestul of your relativesrudele,
127
316000
2000
doar privind cu atenţie la rudele tale,
05:33
but they mayMai be surprisingsurprinzător.
128
318000
3000
dar care pot fi surprinzătoare.
05:36
I did the 23andMeandMe thing
129
321000
2000
Am aplicat metoda 23andMe (23şiEu)
05:38
and was very surpriseduimit to discoverdescoperi that I am fatgras and baldchel.
130
323000
3000
05:41
(LaughterRâs)
131
326000
7000
(Râsete)
05:48
But sometimesuneori you can learnînvăța much more usefulutil things about that.
132
333000
3000
05:51
But mostlyMai ales
133
336000
3000
Dar, în general,
05:54
what you need to know, to find out if you're sickbolnav,
134
339000
2000
05:56
is not your predispositionspredispoziţiile,
135
341000
2000
nu este predispoziţia ta,
05:58
but it's actuallyde fapt what's going on in your bodycorp right now.
136
343000
3000
06:01
So to do that, what you really need to do,
137
346000
2000
Pentru asta, ceea ce trebuie făcut,
06:03
you need to look at the things
138
348000
2000
este să priveşti lucrurile
06:05
that the genesgene are producingproducând
139
350000
2000
pe care genele le produc
06:07
and what's happeninglucru after the geneticsgenetică,
140
352000
2000
şi ce se întâmplă dincolo de genetică.
06:09
and that's what proteomicsproteomica is about.
141
354000
2000
Şi asta înseamnă proteomica.
06:11
Just like genomegenomului mixesAmestecuri the studystudiu of all the genesgene,
142
356000
3000
Aşa cum genomul îmbină studiul tuturor genelor,
06:14
proteomicsproteomica is the studystudiu of all the proteinsproteine.
143
359000
3000
proteomica studiază toate proteinele.
06:17
And the proteinsproteine are all of the little things in your bodycorp
144
362000

146

de fapt maşinile care operează.
06:24
that's where the actionacțiune is.
147
369000
2000
Acolo este toată acţiunea.
06:26
BasicallyPractic, a humanuman bodycorp
148
371000
3000
De fapt, corpul uman
06:29
is a conversationconversaţie going on,
149
374000
3000
este o conversaţie continuă
06:32

150
377000
3000

06:35
and they're tellingspune eachfiecare other to growcrește and to diea muri,
151
380000

06:38
and when you're sickbolnav,
152
383000
2000
Atunci când eşti bolnav,
06:40
something'sceva e goneplecat wronggresit with that conversationconversaţie.
153
385000
2000
ceva nu merge bine în conversaţia asta.
06:42
And so the tricktruc is --
154
387000
2000
Şi şmecheria este --
06:44
unfortunatelydin pacate, we don't have an easyuşor way to measuremăsura these
155
389000
3000
06:47
like we can measuremăsura the genomegenomului.
156
392000
2000
la fel cum facem în cazul genomului.
06:49
So the problemproblemă is that measuringmăsurare --
157
394000
3000
Aşadar, problema este măsurarea --
06:52
if you try to measuremăsura all the proteinsproteine, it's a very elaborateelaborat processproces.
158
397000
3000
06:55
It requiresnecesită hundredssute of stepspași,
159
400000
2000
Necesită sute de paşi,
06:57
and it takes a long, long time.
160
402000
2000
şi durează foarte, foarte mult.
06:59
And it matterschestiuni how much of the proteinproteină it is.
161
404000
2000
Şi contează cantitatea de proteină.
07:01
It could be very significantsemnificativ that a proteinproteină changedschimbat by 10 percentla sută,
162
406000
3000
07:04
so it's not a nicefrumos digitaldigital thing like DNAADN-UL.
163
409000
3000
07:07
And basicallype scurt our problemproblemă is somebody'se cineva in the middlemijloc
164
412000
2000
07:09

165
414000
2000
acestui proces foarte lung,
07:11
they pausepauză for just a momentmoment,
166
416000

se poate opri pentru un simplu moment
07:13
and they leavepărăsi something in an enzymeenzimă for a secondal doilea,
167
418000

07:15
and all of a suddenbrusc all the measurementsmăsurători from then on
168
420000
2000
şi instantaneu toate măsurătorile făcute ulterior
07:17
don't work.
169
422000
2000
nu mai merg.
07:19
And so then people get very inconsistentnepotrivit resultsrezultate
170
424000
2000
07:21
when they do it this way.
171
426000
2000
când este aplicată aceasta metodă.
07:23
People have triedîncercat very hardgreu to do this.
172
428000
2000
07:25
I triedîncercat this a couplecuplu of timesori
173

07:27

07:54
So I was like, "Okay. This guy'sbaieti at leastcel mai puţin resourcefulresurse."

07:56
(LaughterRâs)
187
461000
4000
(Râsete)
08:00
So we starteda început talkingvorbind,
188
465000
2000
Şi am început să discutăm,
08:02
and he said, "I really need a better way to measuremăsura proteinsproteine."
189
467000
3000
08:05
I'm like, "LookedUitat at that. Been there.
190
470000
2000
08:07

191
472000
2000
Nu va fi uşor."
08:09
He's like, "No, no. I really need it.
192
474000
2000
Iar a spus,"Nu, nu. Chiar am nevoie de aşa ceva.
08:11
I mean, I see patientspacienți dyingmoarte everyfiecare day
193
476000
4000
Adică, văd pacienţi murind în fiecare zi
08:15
because we don't know what's going on insideinterior of them.
194
480000
3000
08:18
We have to have a windowfereastră into this."
195
483000
2000
08:20
And he tooka luat me throughprin
196
485000
2000
Şi m-a purtat prin
08:22
specificspecific examplesexemple of when he really neededNecesar it.
197
487000
3000
08:25
And I realizedrealizat, wowWow, this would really make a bigmare differencediferență,
198
490000
2000
08:27
if we could do it,
199
492000
2000
dacă am putea să o facem.
08:29
and so I said, "Well, let's look at it."
200
494000
2000
08:31
AppliedAplicate MindsMintea has enoughdestul playa juca moneybani
201
496000
2000
Applied Minds dispune de bani suficienţi
08:33
that we can go and just work on something
202
498000
2000
08:35
withoutfără gettingobtinerea anybody'soricine e fundingfinanțarea or permissionpermisiune or anything.
203
500000
3000

08:38
So we starteda început playingjoc around with this.
204
503000
2000
08:40
And as we did it, we realizedrealizat this was the basicde bază problemproblemă --
205
505000
3000
08:43
that takingluare the sipSIP of coffeecafea --
206
508000
2000

08:45
that there were humansoameni doing this complicatedcomplicat processproces
207
510000
2000
că oamenii realizează acest proces complicat
08:47
and that what really neededNecesar to be doneTerminat
208
512000
2000
și ceea ce trebuie făcut cu adevărat
08:49
was to automateautomatiza this processproces like an assemblyasamblare linelinia
209
514000
3000

08:54
that would measuremăsura proteomicsproteomica.
211
519000
2000
ce vor măsura proteomica.
08:56
And so we did that,
212
521000
2000
Și așa am făcut.
08:58
and workinglucru with DavidDavid,
213
523000
2000
Lucrând cu David,
09:00
we madefăcut a little companycompanie calleddenumit AppliedAplicate ProteomicsProteomica eventuallyîn cele din urmă,
214
525000
3000
am înfiinţat o companie mică denumită Applied Proteomics (Proteomică Aplicată),
09:03
whichcare makesmărci this roboticrobotizate assemblyasamblare linelinia,
215
528000
3000
care produce această linie de asamblare robotizată,
09:06
whichcare, in a very consistentconsistent way, measuresmăsuri the proteinproteină.
216
531000
3000
care, într-un mod foarte concret, măsoară proteinele.
09:09
And I'll showspectacol you what that proteinproteină measurementmăsurare looksarată like.
217
534000
3000
09:13
BasicallyPractic, what we do
218
538000
2000
De fapt, ceea ce facem
09:15
is we take a dropcădere brusca of bloodsânge
219
540000
2000
este să luăm o picătură de sânge
09:17
out of a patientrabdator,
220
542000
2000
de la un pacient
09:19
and we sortfel out the proteinsproteine
221
544000
2000
și sortăm proteinele
09:21
in the dropcădere brusca of bloodsânge
222
546000
2000
din această picătură de sânge
09:23
accordingin conformitate to how much they weighcântări,
223
548000
2000
conform greutăţii,
09:25
how slipperyalunecos they are,
224
550000
2000
cât de alunecoase sunt
09:27
and we arrangearanja them in an imageimagine.
225
552000
3000
și le aranjăm ca în imaginea afișată.
09:30
And so we can look at literallyliteralmente
226
555000
2000
09:32
hundredssute of thousandsmii of featurescaracteristici at onceo singura data
227
557000
2000
la sute de mii de caracteristici
09:34
out of that dropcădere brusca of bloodsânge.
228
559000
2000
existente în acea picătură de sânge.
09:36
And we can take a differentdiferit one tomorrowMâine,
229
561000
2000
Și putem preleva una diferită mâine,
09:38
and you will see your proteinsproteine tomorrowMâine will be differentdiferit --
230
563000
2000
09:40
they'llei vor be differentdiferit after you eatmânca or after you sleepdormi.
231
565000
3000
09:43
They really tell us what's going on there.
232
568000
3000
09:46
And so this pictureimagine,
233
571000
2000
Această imagine,
09:48
whichcare looksarată like a bigmare smudgePete to you,
234
573000
2000
care arată ca o mare mâzgăleală,
09:50
is actuallyde fapt the thing that got me really thrilledîncântat about this
235
575000
4000
09:54
and madefăcut me feel like we were on the right trackurmări.
236
579000
2000
09:56
So if I zoomzoom into that pictureimagine,
237
581000
2000
Dacă măresc imaginea,
09:58
I can just showspectacol you what it meansmijloace.
238
583000
2000
vă pot arăta exact ce semnifică.
10:00
We sortfel out the proteinsproteine -- from left to right
239
585000
3000
10:03
is the weightgreutate of the fragmentsfragmente that we're gettingobtinerea,
240
588000
3000
este greutatea fragmentelor obținute.
10:06
and from toptop to bottomfund is how slipperyalunecos they are.
241
591000
3000
10:09
So we're zoomingzoom-ul in here just to showspectacol you a little bitpic of it.
242
594000
3000
10:12
And so eachfiecare of these lineslinii
243
597000
2000
Și fiecare dintre aceste linii
10:14
representsreprezintă some signalsemnal that we're gettingobtinerea out of a piecebucată of a proteinproteină.
244
599000
3000
10:17
And you can see how the lineslinii occuravea loc
245
602000
2000
Și puteți observa cum apar liniile
10:19
in these little groupsGrupuri of bumpcucui, bumpcucui, bumpcucui, bumpcucui, bumpcucui.
246
604000
4000
în aceste grupuri mici de pete.
10:23
And that's because we're measuringmăsurare the weightgreutate so preciselyexact that --
247
608000
3000
10:26
carboncarbon comesvine in differentdiferit isotopesizotopi,
248
611000
2000
carbonul are diferiți izotopi,
10:28
so if it has an extrasuplimentar neutronneutroni on it,
249
613000
3000
iar dacă ar avea un neutron in plus,
10:31
we actuallyde fapt measuremăsura it as a differentdiferit chemicalchimic.
250
616000
4000
l-am măsura drept o altă substanță chimică.
10:35
So we're actuallyde fapt measuringmăsurare eachfiecare isotopeizotop as a differentdiferit one.
251
620000
3000
10:38
And so that gives you an ideaidee
252
623000
3000
Așadar, vă puteți da seama
10:41
of how exquisitelySuperb sensitivesensibil this is.
253
626000
2000
cât de extrem de sensibil este.
10:43
So seeingvedere this pictureimagine
254
628000
2000
Privind această imagine
10:45
is sortfel of like gettingobtinerea to be GalileoGalileo
255
630000
2000
este ca și cum am fi Galileo
10:47
and looking at the starsstele
256
632000
2000
privind stelele
10:49
and looking throughprin the telescopetelescop for the first time,
257
634000
2000
prin telescop, pentru prima dată
10:51
and suddenlybrusc you say, "WowWow, it's way more complicatedcomplicat than we thought it was."
258
636000
3000
10:54
But we can see that stuffchestie out there
259
639000
2000
Dar noi, de fapt, putem vedea substanța
10:56
and actuallyde fapt see featurescaracteristici of it.
260
641000
2000
și chiar componentele sale.
10:58
So this is the signaturesemnătură out of whichcare we're tryingîncercat to get patternsmodele.
261
643000
3000
11:01
So what we do with this
262
646000
2000
Ce am face cu acesta
11:03
is, for exampleexemplu, we can look at two patientspacienți,
263
648000
2000
este, de exemplu, să analizăm doi pacienți,
11:05
one that respondeda răspuns to a drugmedicament and one that didn't respondrăspunde to a drugmedicament,
264
650000
3000
11:08
and askcere, "What's going on differentlydiferit
265
653000
2000
și întrebăm, ”Ce este diferit
11:10
insideinterior of them?"
266
655000
2000
în corpul celor doi?”
11:12
And so we can make these measurementsmăsurători preciselyexact enoughdestul
267
657000
3000
Noi putem face măsurătorile suficient de precis
11:15
that we can overlaysuprapunere two patientspacienți and look at the differencesdiferențele.
268
660000
3000
11:18
So here we have AliceAlice in greenverde
269
663000
2000
Aici o avem pe Alice, cu verde,
11:20
and BobBob in redroșu.
270
665000
2000
şi pe Bob, cu roșu.
11:22

Le suprapunem. Acestea sunt date reale.
11:25
And you can see, mostlyMai ales it overlapssuprapuneri and it's yellowgalben,
272
670000
3000
11:28
but there's some things that just AliceAlice has
273
673000
2000
11:30
and some things that just BobBob has.
274
675000
2000
11:32
And if we find a patternmodel of things
275
677000
3000
Dacă am găsi un model al aspectelor
11:35
of the respondersrăspuns to the drugmedicament,
276
680000
3000
pe care le au respondenții la medicament,
11:38
we see that in the bloodsânge,
277
683000
2000
am vedea că în sânge
11:40
they have the conditioncondiție
278
685000
2000
ei au condția
11:42
that allowspermite them to respondrăspunde to this drugmedicament.
279
687000
2000
11:44
We mightar putea not even know what this proteinproteină is,
280
689000
2000
11:46
but we can see it's a markermarker
281
691000
2000
dar putem observa că este un indicator
11:48
for the responseraspuns to the diseaseboală.
282
693000
2000
pentru răspunsul la maladie.
11:53
So this alreadydeja, I think,
283
698000
2000
Cred că acest lucru, este deja
11:55
is tremendouslyturbat usefulutil in all kindstipuri of medicinemedicament.
284
700000
3000
11:58
But I think this is actuallyde fapt
285
703000
2000
Dar cred că este de fapt
12:00
just the beginningînceput
286
705000
2000
doar începutul
12:02
of how we're going to treattrata cancercancer.
287
707000
2000
modului în care vom trata cancerul.
12:04
So let me movemișcare to cancercancer.
288
709000
2000
Acum permiteţi-mi să trec la cancer.
12:06
The thing about cancercancer --
289
711000
2000
Treaba cu cancerul --
12:08
when I got into this,
290
713000
2000
când m-am apucat de asta,
12:10
I really knewștiut nothing about it,
291
715000
2000
chiar nu știam nimic despre el,
12:12
but workinglucru with DavidDavid AgusAgus,
292
717000
2000
dar lucrând cu David Agus,
12:14
I starteda început watchingvizionarea how cancercancer was actuallyde fapt beingfiind treatedtratate
293
719000
3000
12:17
and wenta mers to operationsoperațiuni where it was beingfiind cuta taia out.
294
722000
3000
12:20
And as I lookedprivit at it,
295
725000
2000
Și pe măsură ce priveam,
12:22
to me it didn't make sensesens
296
727000
2000
pentru mine nu avea sens
12:24
how we were approachingse apropie cancercancer,
297
729000
2000
modul în care abordăm cancerul.
12:26
and in orderOrdin to make sensesens of it,
298
731000
3000
Așadar pentru a avea un sens,
12:29
I had to learnînvăța where did this come from.
299
734000
3000
12:32
We're treatingtratare cancercancer almostaproape like it's an infectiousinfecţioase diseaseboală.
300
737000
4000
Tratăm cancerul ca pe orice boală infecțioasă.
12:36
We're treatingtratare it as something that got insideinterior of you
301
741000
2000
12:38
that we have to killucide.
302
743000
2000
și pe care trebuie să îl omorâm.
12:40
So this is the great paradigmparadigmă.
303
745000
2000
Aceasta este marea lecţie.
12:42
This is anothero alta casecaz
304
747000
2000
Acesta este un alt caz
12:44
where a theoreticalteoretic paradigmparadigmă in biologybiologie really workeda lucrat --
305
749000
2000
12:46
was the germgermeni theoryteorie of diseaseboală.
306
751000
3000
teoria microbilor ca sursă a bolilor.
12:49
So what doctorsmedici are mostlyMai ales traineddresat to do
307
754000
2000
12:51
is diagnosediagnostica --
308
756000
2000
este să diagnosticheze --
12:53
that is, put you into a categorycategorie
309
758000
2000
adică să te pună într-o categorie --
12:55
and applyaplica a scientificallyştiinţific provendovedit treatmenttratament
310
760000
2000
12:57
for that diagnosisDiagnosticul --
311
762000
2000
pentru acel diagnostic.
12:59
and that workslucrări great for infectiousinfecţioase diseasesboli.
312
764000
3000
13:02
So if we put you in the categorycategorie
313
767000
2000
13:04
of you've got syphilissifilis, we can give you penicillinpenicilină.
314
769000
3000
sifilisului, îți dăm penicilină.
13:07
We know that that workslucrări.
315
772000
2000
Știm că funcționează.
13:09
If you've got malariamalarie, we give you quininechinină
316
774000
2000
Dacă ai malarie, îți dăm chinină,
13:11
or some derivativederivat of it.
317
776000
2000
sau un derivat al acesteia.
13:13
And so that's the basicde bază thing doctorsmedici are traineddresat to do,
318
778000
3000
Așa sunt instruiți doctorii să facă.
13:16
and it's miraculousmiraculos
319
781000
2000
Și este miraculos
13:18
in the casecaz of infectiousinfecţioase diseaseboală --
320
783000
3000
în cazul bolilor infecțioase --
13:21
how well it workslucrări.
321
786000
2000
cât de bine funcționează.
13:23
And manymulți people in this audiencepublic probablyprobabil wouldn'tnu ar fi be aliveîn viaţă
322
788000
3000
13:26
if doctorsmedici didn't do this.
323
791000
2000
dacă doctorii nu ar fi făcut asta.
13:28
But now let's applyaplica that
324
793000
2000
Să aplicăm această practică
13:30
to systemssisteme diseasesboli like cancercancer.
325
795000
2000
la bolile de sistem, precum cancerul.
13:32
The problemproblemă is that, in cancercancer,
326
797000
2000
Problema este că, la cancer,
13:34
there isn't something elsealtfel
327
799000
2000
nu este altceva
13:36
that's insideinterior of you.
328
801000
2000
în corpul tău.
13:38
It's you; you're brokenspart.
329
803000
2000
Tu ești, tu ești defect.
13:40
That conversationconversaţie insideinterior of you
330
805000
4000
Conversația din interiorul tău
13:44
got mixedamestecat up in some way.
331
809000
2000
s-a defectat într-un fel.
13:46
So how do we diagnosediagnostica that conversationconversaţie?
332
811000
2000
Cum diagnosticăm acea conversație?
13:48
Well, right now what we do is we dividedivide it by partparte of the bodycorp --
333
813000
3000
13:51
you know, where did it appearapărea? --
334
816000
3000
adică acolo unde au apărut prima dată --
13:54
and we put you in differentdiferit categoriescategorii
335
819000
2000
și te punem în categorii diferite
13:56
accordingin conformitate to the partparte of the bodycorp.
336
821000
2000
în funcție de partea corpului.
13:58
And then we do a clinicalclinic trialproces
337
823000
2000
Apoi facem un test clinic
14:00
for a drugmedicament for lungplămân cancercancer
338
825000
2000
pentru un medicament împotriva cancerului la plamâni
14:02
and one for prostateprostată cancercancer and one for breastsân cancercancer,
339
827000
3000
14:05
and we treattrata these as if they're separatesepara diseasesboli
340
830000
3000
14:08
and that this way of dividingdespărțitor them
341
833000
2000
și acest mod de a le clasifica
14:10
had something to do with what actuallyde fapt wenta mers wronggresit.
342
835000
2000
14:12
And of coursecurs, it really doesn't have that much to do
343
837000
2000
Bineînțeles, nu prea are multă legătură
14:14
with what wenta mers wronggresit
344
839000
2000
cu ceea ce nu funcționează bine.
14:16
because cancercancer is a failureeșec of the systemsistem.
345
841000
3000
Deoarece cancerul este o defecțiune a sistemului.
14:19
And in factfapt, I think we're even wronggresit
346
844000
2000
14:21
when we talk about cancercancer as a thing.
347
846000
3000
vorbim despre cancer ca despre un lucru.
14:24
I think this is the bigmare mistakegreşeală.
348
849000
2000
Cred că este o mare greșeală.
14:26
I think cancercancer should not be a nounsubstantiv.
349
851000
4000
14:30
We should talk about canceringCancering
350
855000
2000
Ar trebui să discutăm despre cancerizare
14:32
as something we do, not something we have.
351
857000
3000
14:35
And so those tumorstumori,
352
860000
2000
Astfel, aceste tumori,
14:37
those are symptomssimptome of cancercancer.
353
862000
2000
sunt simptomele cancerului.
14:39
And so your bodycorp is probablyprobabil canceringCancering all the time,
354
864000
3000
Corpul tău probabil cancerizează tot timpul.
14:42
but there are lots of systemssisteme in your bodycorp
355
867000
3000
14:45
that keep it undersub controlControl.
356
870000
2000
care îl țin sub control.
14:47
And so to give you an ideaidee
357
872000
2000
Pentru a vă oferi o idee
14:49
of an analogyanalogie of what I mean
358
874000
2000
despre analogia pe care o fac
14:51
by thinkinggândire of canceringCancering as a verbverb,
359
876000
3000
gândindu-ne la cancerizare ca un verb,
14:54
imagineimagina we didn't know anything about plumbinginstalatii sanitare,
360
879000
3000
imaginați-vă că nu știm absolut nimic despre instalații
14:57
and the way that we talkeda vorbit about it,
361
882000
2000
și modul în care discutăm despre ele,
14:59
we'dne-am come home and we'dne-am find a leakspărtură in our kitchenbucătărie
362
884000
3000
15:02
and we'dne-am say, "Oh, my housecasă has waterapă."
363
887000
4000
şi spunem, ”Ah, casa mea are apă.”
15:06
We mightar putea dividedivide it -- the plumberinstalator would say, "Well, where'sîn cazul în care este the waterapă?"
364
891000
3000
Clasificăm problema -- instalatorul ar spune ”E bine, unde este apa?”
15:09
"Well, it's in the kitchenbucătărie." "Oh, you musttrebuie sa have kitchenbucătărie waterapă."
365
894000
3000
”Păi, este în bucătărie.” ”Ah, atunci aveți apă de bucătărie.”
15:12
That's kinddrăguț of the levelnivel at whichcare it is.
366
897000
3000
Cam la acest nivel vorbim.
15:15
"KitchenBucatarie waterapă,
367
900000
2000
”Apă de bucătărie?
15:17
well, first of all, we'llbine go in there and we'llbine mopmop out a lot of it.
368
902000
2000
15:19
And then we know that if we sprinklese presara DranoDrano around the kitchenbucătărie,
369
904000
3000
15:22
that helpsajută.
370
907000
3000
ne va ajuta.
15:25
WhereasÎntrucât livingviaţă roomcameră waterapă,
371
910000
2000
Când vine vorba despre apa din sufragerie,
15:27
it's better to do targudron on the roofacoperiş."
372
912000
2000
este mai bine sa gudronăm acoperișul.”
15:29
And it soundssunete sillyprost,
373
914000
2000
Şi sună prostește,
15:31
but that's basicallype scurt what we do.
374
916000
2000
15:33
And I'm not sayingzicală you shouldn'tnu ar trebui mopmop up your waterapă if you have cancercancer,
375
918000
3000
Nu spun că nu trebuie curățată apa dacă ai cancer.
15:36
but I'm sayingzicală that's not really the problemproblemă;
376
921000
3000
Dar spun că nu aceasta este problema cu adevărat;
15:39
that's the symptomsimptom of the problemproblemă.
377
924000
2000
ci doar simptomul problemei.
15:41
What we really need to get at
378
926000
2000
Trebuie, în realitate, să ajungem la
15:43
is the processproces that's going on,
379
928000
2000
procesul care se desfășoară
15:45
and that's happeninglucru at the levelnivel
380
930000
2000
și care are loc la nivelul
15:47
of the proteonomicproteonomic actionsacţiuni,
381
932000
2000
acțiunii proteonomice,
15:49
happeninglucru at the levelnivel of why is your bodycorp not healingcicatrizare itselfîn sine
382
934000
3000
15:52
in the way that it normallyîn mod normal does?
383
937000
2000
aşa cum face în mod normal?
15:54
Because normallyîn mod normal, your bodycorp is dealingcare se ocupă with this problemproblemă all the time.
384
939000
3000
15:57
So your housecasă is dealingcare se ocupă with leaksscurgeri all the time,
385
942000
3000
16:00
but it's fixingfixare them. It's drainingdrenarea them out and so on.
386
945000
4000
Dar le repară. Le drenează și tot așa.
16:04
So what we need
387
949000
3000
În concluzie, ceea ce este necesar
16:07
is to have a causativecauzal modelmodel
388
952000
4000
este să avem un model cauzal
16:11
of what's actuallyde fapt going on,
389
956000
2000
a ceea ce se întâmplă în realitate.
16:13
and proteomicsproteomica actuallyde fapt gives us
390
958000
3000
Proteomica ne oferă de fapt
16:16
the abilityabilitate to buildconstrui a modelmodel like that.
391
961000
3000
16:19
DavidDavid got me inviteda invitat
392
964000
2000
David mi-a obţinut o invitaţie
16:21
to give a talk at NationalNaţionale CancerCancer InstituteInstitutul
393
966000
2000
16:23
and AnnaAnna BarkerBarker was there.
394
968000
3000
şi Anna Barker era acolo.
16:27
And so I gavea dat this talk
395
972000
2000
Am susținut această prelegere
16:29
and said, "Why don't you guys do this?"
396
974000
3000
16:32
And AnnaAnna said,
397
977000
2000
Anna a răspuns,
16:34
"Because nobodynimeni withinîn cancercancer
398
979000
3000
”Deoarece nimeni din domeniul cancerului
16:37
would look at it this way.
399
982000
2000
nu ar privi lucrurile din aceasta perspectiva.
16:39
But what we're going to do, is we're going to createcrea a programprogram
400
984000
3000
16:42
for people outsidein afara the fieldcamp of cancercancer
401
987000
2000
pentru persoanele din afara domeniului cancerului,
16:44
to get togetherîmpreună with doctorsmedici
402
989000
2000
care să facă echipă cu doctorii
16:46

16:49
and work out differentdiferit programsprograme of researchcercetare."
404
994000

16:53
So DavidDavid and I appliedaplicat to this programprogram

that will last for fivecinci yearsani
414
1020000
2000
care va dura cinci ani,
17:17
where we're really going to try to buildconstrui a modelmodel of cancercancer like this.
415
1022000
3000
17:20
We're doing it in micesoareci first,
416
1025000
2000
Pentru început vom face teste pe șoareci.
17:22
and we will killucide a lot of micesoareci
417
1027000
2000
Vom omorî foarte mulți șoareci
17:24
in the processproces of doing this,
418
1029000
2000
în procesul de cercetare,
17:26
but they will diea muri for a good causecauza.
419
1031000
2000
dar vor muri pentru cauză bună.
17:28
And we will actuallyde fapt try to get to the pointpunct
420
1033000
3000
Astfel, încercăm să ajungem în punctul
17:31
where we have a predictivecu funcþia de predicþie modelmodel
421
1036000
2000
în care vom obține un model predictiv
17:33
where we can understanda intelege,
422
1038000
2000
pe baza căruia putem înțelege
17:35
when cancercancer happensse întâmplă,
423
1040000
2000
când apare cancerul,
17:37
what's actuallyde fapt happeninglucru in there
424
1042000
2000
ce se întâmplă acolo
17:39
and whichcare treatmenttratament will treattrata that cancercancer.
425
1044000
3000
și ce tratament este necesar pentru vindecare.
17:42
So let me just endSfârşit with givingoferindu- you a little pictureimagine
426
1047000
3000
În final, voi încheia prezentându-vă o imagine
17:45
of what I think cancercancer treatmenttratament will be like in the futureviitor.
427
1050000
3000
17:48
So I think eventuallyîn cele din urmă,
428
1053000
2000
Cred că într-un final,
17:50
onceo singura data we have one of these modelsmodele for people,
429
1055000
2000
odată ce avem aceste modele pentru oameni,
17:52
whichcare we'llbine get eventuallyîn cele din urmă --
430
1057000
2000
pe care le vom obţine într-un final --
17:54
I mean, our groupgrup won'tnu va get all the way there --
431
1059000
2000
17:56
but eventuallyîn cele din urmă we'llbine have a very good computercomputer modelmodel --
432
1061000
3000
17:59
sortfel of like a globalglobal climateclimat modelmodel for weathervreme.
433
1064000
3000
18:02
It has lots of differentdiferit informationinformație
434
1067000
3000
Conține foarte multe informații diferite
18:05
about what's the processproces going on in this proteomicproteomic conversationconversaţie
435
1070000
3000
18:08
on manymulți differentdiferit scalescântare.
436
1073000
2000
la multe scale diferite.
18:10
And so we will simulatesimula
437
1075000
2000
Vom simula
18:12
in that modelmodel
438
1077000
2000
în acel model
18:14
for your particularspecial cancercancer --
439
1079000
3000
pentru cancerul tău specific --
18:17
and this alsode asemenea will be for ALSALS,
440
1082000
2000
la fel şi pentru scleroză amiotrofică laterală
18:19
or any kinddrăguț of systemsistem neurodegenerativeneurodegenerative diseasesboli,
441
1084000
3000
sau orice altă maladie neurodegenerativă a sistemului,
18:22
things like that --
442
1087000
2000
boli similare --
18:24
we will simulatesimula
443
1089000
2000
vom simula
18:26
specificallyspecific you,
444
1091000
2000
doar pentru tine, în mod specific,
18:28
not just a genericgeneric personpersoană,
445
1093000
2000
nu doar pentru o persoană generică,
18:30
but what's actuallyde fapt going on insideinterior you.
446
1095000
2000
18:32
And in that simulationsimulare, what we could do
447
1097000
2000
În această simulare, ceea ce putem face
18:34
is designproiecta for you specificallyspecific
448
1099000
2000
este să proiectăm specific pentru tine
18:36
a sequencesecvenţă of treatmentstratamente,
449
1101000
2000
o secvență de tratamente,
18:38
and it mightar putea be very gentleblând treatmentstratamente, very smallmic amountssume of drugsdroguri.
450
1103000
3000
18:41
It mightar putea be things like, don't eatmânca that day,
451
1106000
3000
18:44
or give them a little chemotherapychimioterapie,
452
1109000
2000
sau să le facem o mică chimioterapie,
18:46
maybe a little radiationradiație.
453
1111000
2000
sau poate o mică iradiere.
18:48
Of coursecurs, we'llbine do surgeryinterventie chirurgicala sometimesuneori and so on.
454
1113000
3000
18:51
But designproiecta a programprogram of treatmentstratamente specificallyspecific for you
455
1116000
3000
18:54
and help your bodycorp
456
1119000
3000
și oferind ajutor corpului tău
18:57
guideghid back to healthsănătate --
457
1122000
3000
să revină la starea de sănătate --
19:00
guideghid your bodycorp back to healthsănătate.
458
1125000
2000
îți vei ghida corpul să redevină sănătos.
19:02
Because your bodycorp will do mostcel mai of the work of fixingfixare it
459
1127000
4000
19:06
if we just sortfel of proppropunerii it up in the waysmoduri that are wronggresit.
460
1131000
3000
19:09
We put it in the equivalentechivalent of splintsatele.
461
1134000
2000
Ceva similar cu atelele.
19:11
And so your bodycorp basicallype scurt has lots and lots of mechanismsmecanisme
462
1136000
2000
Corpul uman dispune de foarte multe mecanisme
19:13
for fixingfixare cancercancer,
463
1138000
2000
pentru tratarea cancerului,
19:15
and we just have to proppropunerii those up in the right way
464
1140000
3000
19:18
and get them to do the jobloc de munca.
465
1143000
2000
și să le punem la treabă.
19:20
And so I believe that this will be the way
466
1145000
2000
Cred că aceasta va fi metoda
19:22
that cancercancer will be treatedtratate in the futureviitor.
467
1147000
2000
19:24
It's going to requirenecesita a lot of work,
468
1149000
2000
Va necesita foarte multă muncă,
19:26
a lot of researchcercetare.
469
1151000
2000
multă cercetare.
19:28
There will be manymulți teamsechipe like our teamechipă
470
1153000
3000
19:31
that work on this.
471
1156000
2000
care vor lucra la asta.
19:33
But I think eventuallyîn cele din urmă,
472
1158000
2000
Dar, într-un final, cred
19:35
we will designproiecta for everybodytoata lumea
473
1160000
2000
că vom proiecta pentru toată lumea
19:37
a custompersonalizat treatmenttratament for cancercancer.
474
1162000
4000
un tratament personalizat pentru cancer.
19:41
So thank you very much.
475

Vă mulțumesc foarte mult.
